Luciferase and GFP expressing Amplicon vectors were stereotactically inoculated in the hippocampus of mouse brain.

Luciferase (In Vivo Imaging System) and GFP (native fluorescence) expressions were monitored over time, up to 6 months when the experiment was terminated.

GFP positive cells, including cell bodies and projecting axons, were characterized as neurons.

Long term and specific expression of the luciferase and GFP genes was obtained in neurons following intra-hippocampal injection of Amplicon vectors.

HSV amplicon are powerful vectors for long term gene expression in vivo:

 

-  High efficiency of gene transfer

-  Strong efficacy of gene expression

-  Long term expression in neurons

-  Amplicon vectors are safe (no cytotoxicity)

-  Amplicon vectors are easy to manipulate

-  Amplicon vectors can be produced at high scale
